Проведен скрининг нуклеотидных последовательностей гена fliC к сальмонеллам различных видов, циркулирующих у свиней, пушных зверей, птиц и КРС, и подобраны специфичные праймеры к Salmonella typhimurium. В результате подбора оптимальных концентраций реакционных смесей и параметров амплификации сконструирована тест-система ПЦР для детекции генома Salmonella typhimurium. Она обладает специфичностью, отсутствием неспецифических шмеров, чувствительностью не менее 20 копий геномов ДНК в исследуемом образце.
The screening of the nucleotide sequences of the fliC gene to salmonella of various species circulating in pigs, fur animals, birds and cattle was carried out and specific primers for Salmonella typhimurium were selected. As a result of selection of optimal concentrations of reaction mixtures and filtration parameters, a PCR test system was designed to detect the genome of Salmonella typhimurium. It has specificity, the absence of nonspecific Schmer, sensitivity of at least 20 copies of DNA genomes in the sample.
